  • While Osmosis's backstory is laced with humor, the subtext of it is kind of disheartening. After she lost her mother, Shane entered into a school science fair and began to come out of her shell. But after Frank ate some bad oysters at the fair, it left Osmosis between a rock and a hard place: either alert backup and/or security and wait for them while letting Frank potentially get fatally sick, or expel the virus immediately and likely cause an embarrassing situation for Frank in front of everyone. The ending of the backstory acts sort of like an In-Universe Bittersweet Ending: while Osmosis's choice did save Frank, it led to both of them being made into public laughing stocks (with Frank additionally having a restraining order put on him and Osmosis additionally being discredited in the eyes of his peers and superiors), landed them both in bad situations employment-wise (Frank was fired from his job at the local pea-soup factory and subsequently forced to take a lower-paying one at the zoo, while Osmosis was suspended for "unnecessary force" and subsequently re-assigned to patrol duty in the mouth), and left poor Shane more introverted than ever before.
    Osmosis: Since then not a day has gone by that I'd have to I wonder "Did I do the right thing?"
  • Drix's Heroic BSoD after the Mayor tells him he's only but a hyped-up cold medicine for 'temporary relief'.
  • The Mayor firing Jones and Drix. Right after Jones and Drix supposedly defeated Thrax, the Mayor is more concerned over the damage Jones caused. After Jones makes a comment how the Mayor is more concerned over his campaign than everyone's lives, the Mayor fires him on the spot. Everyone in the room, even Leah and the Police Chief are left distraught; despite their views on Jones and his actions, even they believe this was a bit harsh. Jones said it best:
    Osmosis: Figures, I finally do something right for Frank, and I get fired for it.
    • Drix even tries to stand up for Jones, but the Mayor is already fed up with the situation and brings up Drix's temporary contract and has him deported.
    • Even worse, when Jones is trudging the streets afterwards in the rain, he sees a newspaper drop off in front of a corner store. The headline reads "LOSER COP FIRED", complete with a picture of Jones front and center, to the point that he shies away from the area to prevent the vendor from recognizing him. From this, it becomes apparent that the Mayor ensured that the press got wind of Jones' termination and reminded them of his past "record" to ensure that no one would believe or listen to him if he tried to speak out against the Mayor's policies or reveal Thrax's presence to the public.
  • Once the theft of Frank's hypothalamus starts becoming apparent, he passes out while driving. Uncle Bob, who's been nothing but pure comic relief prior to this point, is extremely alarmed and actually bursts into tears once he sees Shane after Frank is loaded into an ambulance.
  • We cut to the inside of Frank's body as he approaches near-death, and it not pretty: the city of Frank is in chaos, the ordinarily stoic anchorman is driven to angry despair, a group of rich cells decides to pull a While Rome Burns and play their violins when faced with imminent death, Leah and Drix are crestfallen over Ozzy (seemingly) not making it, and even Mayor Phlemming, seeing his city collapsing into the ground, is remorseful over what he's done.
  • When Shane hears her father's heartbeat flatline, she runs back into the E.R. in total heartbreak, thinking she's about to lose the only parent she has left. She cries her heart out in apology, taking back what she said the last time they spoke, saying she thinks Frank is the greatest dad in the world. You can truly see just how upset Shane is by thinking the last exchange they'll ever have is a fight.
    • Even more, it's possible she might've been afraid that her words might've caused her father to lose the will to keep living, as he is shown throughout the film to still be battling depression brought on by her mother's death, and in her eyes, their fight might have been the catalyst for him to finally give up.
    • Just look at the doctors and nurses in the room, even though they probably seen this happen loads of time, it is clear that it never gets easier for them to see this.
    • Miraculously, thanks to Ozzy, this turns into a Tear Dryer as just as the doctor prepares to sign off on Frank's death certificate with the Time Of Death...

    Nurse: Wait a minute! He's got a pulse!
    Doctor: He's coming back! 107.7, 107.6...
    Frank: Your Momma says 'hi.'
