  • James Brown is the Reverend Cleophus James, the gospel singing preacher who shows Jake the light and inspires him to get the band back together.
  • Cab Calloway is the father figure and mentor to the brothers who introduced them to their suits and delivers a barnstormer of "Minnie the Moocher".
  • Ray Charles is the blind owner of Ray's Music Exchange, where the band picks up their instruments.
  • Aretha Franklin is Matt Murphy's hard-pressed wife at the soul food diner.
  • Steve "the Colonel" Cropper steps in as the band's lead guitar, a member of Murph's Magic Tones.
  • Donald "Duck" Dunn, another member of the Magic Tones, is the band's bassist.
  • George "Murphy" Dunn plays keyboards and was the lead singer of the Magic Tones.
  • Willie "Too Big" Hall is the band's drummer, from the Magic Tones.
  • Tom "Bones" Malone, an SNL band member and Blues Brother legend, is the band's saxophonist.
  • Matt "Guitar" Murphy, Aretha's henpecked husband, is another of the band's lead guitarists.
  • Lou Marini jumps in on the trombone.
  • "Mr. Fabulous" Alan Rubin, the maitre d'hotel at the French restaurant, rejoins the band as their trumpet player.
  • Carrie Fisher is the Mystery Woman, Jake's estranged wife who keeps trying to kill him.
  • Henry Gibson is the lead Nazi.
  • John Candy joins the show as Burt Mercer, the lead detective chasing the brothers.
  • John Lee Hooker gives us a street show outside of Ray's Music Exchange.
  • Kathleen Freeman plays an impossible magical nun, mother figure to the brothers.
  • Steve Lawrence shows up to criticize the brothers for still wearing their verkakte suits and come back in as their manager.
  • Twiggy steps in at a gas station so Elwood can hit on her and ditch her.
  • Frank Oz is the prison guard who returns Jake's possessions (including a soiled prophylactic).
  • Jeff Morris is Bob, the owner of Bob's Country Bunker.
  • Character actor Charles Napier is the lead singer and Winnebago driver for the Good Ol' Boys, McElroy.
  • Steven Spielberg is the Cook County Assessor who takes the brothers' check at the end of the movie.
  • Descended Creator John Landis is one of the cops who chases the brothers through the mall.
  • Joe Walsh is one of the prison inmates.
  • Paul Reubens is a Retroactive Recognition cameo as a waiter at, ironically, Mr. Fabulous Alan Rubin's French restaurant.
  • SNL writer and actor Rosie Shuster is one of the waitresses at Aretha's diner.
  • Chaka Khan is one of the choir at Cleophus James's church.
  • Layne "Shotgun" Britton, a well-connected character actor is the guy who asks if Elwood remembered to grab him some Cheez-Wiz.
  • Pinetop Perkins, a blues musician, plays alongside John Lee Hooker and argues with him over who wrote "Boom Boom".
  • Carolyn Franklin, Aretha's younger sister and a singer-songwriter in her own right, is one of the singers at the diner.
  • De'Voureaux White, aka Argyle the limo driver is the kid who tries to steal a guitar from Ray's, another bit of Retroactive Recognition.
  • James Avery (Uncle Phil) has an uncredited role as one of the detectives after the brothers. More Retroactive Recognition.
  • Character actor Ralph Foody is the police dispatcher who comments on the brothers' antics in the third act. He says, among other things, that the use of unnecessary violence in the apprehension of the Blues Brothers has been approved.
