- Acting for Two: Cree Summer pulls triple duty as Mercy Graves, Livewire, and Mad Harriet.
- All-Star Cast: The game features a lot of the same talent as the previous game, but also has names like John Barrowman, Michael Ironside, Michael Dorn, and Mark Hamill all reprising their roles, and several more actors that were from the DCAU, like Kevin Conroy, Michael Rosenbaum, and Susan Eisenberg.

- When Harley uses her musical ability, you can play the song "Hit Me With Your Best Shot" by Pat Benatar.
- At the disco diner in Metropolis, you can jam to "One Way or Another" by Blondie.
- Role Reprise: A significant number of voice actors reprise their roles from other incarnations of the characters.
- Like in LEGO Batman 3, we have Clancy Brown as Lex Luthor, Tara Strong as Harley Quinn, Josh Keaton as Hal Jordan, Bumper Robinson as Cyborg, Travis Willingham as Superman & Booster Gold, Roger Craig Smith as Skeets, JB Blanc as Penguin, Fred Tatasciore as Solomon Grundy, Steven Jay Blum as Captain Cold, Erica Luttrell as Cheetah, Gilbert Gottfried as Mr. Mxyzptlk, as well as Marc Worden and Ike Amadi coming back from the third game in the respective roles of Sinestro and Atrocitus.

- Reprising from Superman/Shazam!: The Return of Black Adam: Zach Callison as Billy Batson.
- Reprising from Justice League: Doom in addition to Batman, Wonder Woman, and Cyborg: Michael Rosenbaum as the Flash (given he's Barry, not Wally).

- Reprising from Superman vs. the Elite: Fred Tatasciore as Perry White.
- Reprising from LEGO DC Comics Super Heroes: Justice League — Attack of the Legion of Doom in addition to Green Lantern: Mark Hamill as the Trickster (as this Trickster is Axel Walker, not James Jesse like in The Flash (1990), the DCAU, and the Arrowverse).
- Reprising from the DC Extended Universe: Zachary Levi as Shazam (Movie).
- Averted with Steve Blum as Count Vertigo as the design used belongs to New 52's Werner Zytle, not Werner Vertigo like in DC Showcase: Green Arrow or Young Justice (2010); Jason Marsden as the Atom as he's voicing Ryan Choi as opposed to Ray Palmer like in YJ or the DC Nation Sword of the Atom shorts; and Jennifer Hale as Killer Frost as she voiced the Louise Lincoln incarnation in the DC Animated Universe, but the character bio shows that this game's Killer Frost is the Caitlin Snow incarnation.
- Among several reprises in the Latin American dub, two stand out due to the significant amount of time since their last performance as their characters. José Luis Orozco returns as Two-Face after voicing him in Batman (1989) and Batman Forever, whereas Rubén Moya voices Darkseid for the first time since Super Friends: The Legendary Super Powers Show, over 30 years before.
- Show Accuracy/Toy Accuracy: The game notably veers away from toy accuracy and toward show accuracy, a rarity for the LEGO games. Of the characters who have real-world minifigs, very few actually resemble the toys.
- Subverted by the fact that many of the Minifigures that were made after the game's release are nigh-identical to their appearance in-game. Whether the designs in the game were based on then-unreleased Minifigures or if the figures in the 2019 waves were based on the game's designs is unknown, however.
