Like GMod Tower, Tower Unite is loaded to the brim with references to other media.

General
- An unlockable cosmetic item for the player is a white and blue soda drink hat with a red #1 etched on it. Additionally, the diving helmet's description is "Who lives in a pineapple under the sea?". The pineapple tube's description takes this a bit further by saying "It floats, so you won't see it under the sea anytime soon".
- During Halloween, one of the gravestones in the plaza reads "Here lies MacDGuy's hopes and dreams".note
- The Stripe Hat and Iridescent Hat are clear references to The Cat in the Hat and Back to the Future.
- The fedora's description is "Don't forget to grab it before the door closes."
- The Shark tube's description is "Fish are friends. Not Food.", and the Racecar tube's description is "I am speed".
- The Old Candelabra item's description ends with "Note: Does not talk."
- Virtually any Mushroom based item's description will reference Super Mario Bros. in some form.
- The Barrel item's description says it's "Slightly worn from being thrown a lot."
- The fairy pet, which consists of a bright ball with wings, has a description that reads "Hey, listen!". The bottle item's description is also "Sadly, it does not contain a fairy in it".
- The Cardboard Box item's description says it's "A snake's favorite hiding spot."
- The throwable blood wine item, complete with the description "What is a catsack? A miserable little pile of secrets!"
- The description for the Falling Money particle effect (A Casino milestone) reads "Make it rain lods e mone."
- A milestone for the Typing Derby is a Throwable Keyboard, with its description reading: "Sometimes you just gotta rage it out". This can easily bring to mind a certain individual known for doing exactly that with his keyboard.
- The Blender item's description reads "Blends just about anything, but will it?". The "Let's Blend!" logo on it even bears a strong resemblance to the videos its description references.
- The Hot Tub's description simply reads "WooHoo!" It's spelled exactly like a certain mechanic in The Sims where Sims can "get busy", including the very item the description is on.
- As of version 0.10.2.0, the Wowozela from the arcade is capable of emitting A single note by Hatsune Miku, Morshu's signature "Mmmmmm!", and the sound used by the "Skull Trumpet" video. Makes it worth finally getting considering it costs 2.5 million tickets.
- A non-item example: Rob, the shopkeeper of Rob's Imports, is the only shopkeeper not physically present and is instead just reached via a phone. Sounds pretty similar to the father of a certain boy with PSI powers.
- In the Arcade, there are three third-party plushies you can earn either via the Pluck-a-Pal crane game or from the prize counter, those being the Zeeky H. Bomb and Kamikaze Watermelon, as well as an Albino Blacksheep.
- The Tower Glove, a quality of life item introduced in version 0.15.0, is very much based on Nintendo's Power Glove.
- Two in the Chalkboard furniture item: the description reads "Rudy's got the chalk.", while text on the actual chalkboard reads "You can spin me, that's a neat trick!".
- The Typing Derby takes excerpts from a lot of existing media that are randomly chosen every time a new game is started on them. Some notable ones are listed below (some of which are believed to be new to the game as of version 0.17.3.0 in 2023):
- The first half of "Shia LaBeouf" Live.
- The Tragedy of Darth Plagueis the Wise.
- Yoshikage Kira's Cold Ham Motive Rant.
