After the events of Jurassic Park, Simon Masrani, the founder of the Masrani Global Corporation bought out InGen in 1997 after the death of John Hammond, and promised on Hammond's deathbed that he could make his dream of a dinosaur theme park come true. In 2005, the park, now named Jurassic World was finally opened to the public. The park has thrived for a decade without incidents, causing the staff of the park to become complacent in their success. With the cloning of dinosaurs in the 90's now common knowledge that is covered in science textbooks, the general public is losing interest in Jurassic World. CEO Simon Masrani has ordered the development of a genetically-forged hybrid dinosaur in order to recapture the public's interest. Dr. Henry Wu, a hold-over from the days of Jurassic Park, has engineered the Indominus rex, a large carnivore made from a classified list of various species of dinosaurs and modern-day animals.

Zach and Gray, a pair of brothers, arrive on the island around Christmastime to visit the park, as their aunt Claire Dearing is the park operations manager of Jurassic World. Upon arrival they are escorted to Claire by her hapless assistant Zara Young, who is less than thrilled with the prospect of babysitting the boys at the park, as Claire is unable to bring herself to leave her job. The boys eventually ditch poor Zara and go off to explore the bustling park on their own. During their tour, Gray reveals to Zach that their parents are getting divorced, and Zach gives his little brother some Tough Love counseling if there ever was one.

Masrani and Claire go to the Indominus' paddock to inspect the new dinosaur, and the former is concerned that the beast has already attempted to escape and kill its handlers numerous times. Masrani asks Claire to bring in Owen Grady, a former Navy man who has raised a pack of Velociraptors - Blue, Delta, Echo, and Charlie - from birth and has established himself as the alpha of their pack, in order to inspect the paddock. Owen and his partner Barry find themselves regularly harassed by InGen's head of security Vic Hoskins, who wishes to one day weaponize dinosaurs after seeing Owen's relationship with the raptors first hand. Owen and Barry insist that the raptors are still dangerous wild animals, but their arguments fall on deaf ears.

After some tense banter between the two - the pair having gone on a failed date in the past - Claire and Owen return to the Indominus paddock. Owen immediately begins to notice several flaws in the treatment of the Indominus: the dinosaur was raised in a paddock that was too small for it all of its' life, she was raised in isolation (having eaten her sibling when it was young), and has not been allowed to hunt and hone her natural instinct. Claire's frustration turns to horror when claw marks are seen going to the top of the paddock's walls, indicating that the hybrid has escaped. Owen and two workers enter the paddock to investigate, and Claire calls control room workers Lowery and Vivian to check for the Indominus' tracking chip. Lowery claims the Indominus is still in her paddock and that there are people inside it, to Claire's horror. Owen and the workers are alerted, and realize too late the Indominus has faked its escape in order to stage its actual escape. The two workers are killed, inadvertently allowing the Indominus to escape.

Owen arrives in the control room, where Masrani refuses to put the park on alert, believing that the island's Asset Containment Unit can capture the creature. Despite Owen's protests, the ACU unit moves in to confront the Indominus, who reveals its ability to camouflage and ambushes them, killing most of them since Masrani denied the use of lethal weapons on the expensive animal. Owen insists they need to put the creature down before it does any more damage, and pleads Claire to evacuate the island; she eventually decides to move the park's visitors back to the central area and begin the evacuation. Masrani confronts Wu, and though Wu refuses to reveal the component parts of the Indominus, admits its' camouflage came from cuttlefish and it gained the ability to hide from thermal radiation from tree frog DNA. Wu throws Masrani's frustration back in his face, clarifying that he has only done what Masrani ordered.

Zach and Gray enter the park's gyrosphere ride just as the evacuation order arrives, and the boys go off-road to explore. They find themselves in danger when the Indominus attacks them and a small herd of Ankylosaurus, and narrowly escape the Indominus by leaping into a river. Claire recruits Owen to help find her nephews in the gyrosphere valley, only to find that the Indominus is massacring the park's dinosaurs for sport. Zach and Gray venture into the island's restricted zone, where they find the remains of the old Jurassic Park visitor center. The boys manage to repair an old jeep and use it to return to the main park.

Hoskins appeals to Masrani to use the raptor pack to hunt down the Indominus, but Masrani staunchly refuses to release such dangerous animals onto the island. Masrani follows Owen's earlier advice and has a mini-gun planted in a helicopter, and personally flies some ACU troops with him into battle. They find the Indominus and try to kill it, only for the hybrid to break into the park's pterosaur aviary. The pterosaurs are scared out of their home by the Indominus and quickly attack the helicopter, causing it to crash and kill Masrani and the troopers. The freed pterosaurs take their frustrations out by attacking a virtual buffet of visitors who have been herded into Main Street, just as Zach and Gray return. Zara tries to save Zach and Gray, but ends up dying a horrible death after being fought over by several pterosaurs and ultimately getting eaten by the park's Mosasaurus. Owen, Claire, and the ACU arrive to fight the swarm of pterosaurs, and Claire ends up saving Owen from a Dimorphodon, and having won Owen over, shares a kiss with him. Claire reunites with her nephews and the group retreats from the overrun Main Street.

With the death of Masrani, Hoskins assumes command of Isla Nublar and goes to prepare Owen's pack for a hunt. Despite his reservations, Owen is convinced to use his raptors to hunt down the Indominus. Owen, Barry, a group of InGen mercenaries hired by Hoskins, and the raptor pack find the Indominus, but the hybrid reveals its' raptor genes when it communicates with the pack, and convinces them to become her underlings and turn on the humans. Most of Hoskins' mercenaries are killed, as is the raptor Charlie, while Owen, Barry, Claire, and the boys narrowly escape the pack.

As the park's visitors are slowly evacuated, the staff of Jurassic World's control room begins to leave as well. Lowery bravely decides to stay behind to help Claire until she gets out, and tries to dramatically kiss Vivian... only for his coworker to reveal she has a boyfriend she's never mentioned at work. Awkward.

Owen, Claire and the boys head for the helicopter pad to await their escape, but Claire notices that Wu's genetics lab has been cleared out. Hoskins arrives and reveals that he and Wu have made a deal to pit the Indominus and the raptors against each other, planning to sell the genomes of the winner to military forces. Hoskins admits the Indominus needs to be scaled down, but is suddenly killed by Delta, a raptor who antagonized Hoskins. The surviving raptor pack corner the group, but they are convinced to fight the Indominus to save Owen after he reestablishes their bond. Blue, the pack's beta, is knocked out, leaving Owen, Delta, and Echo to battle the Indominus. As the fight begins to go south for Owen's pack as Delta and Echo are killed, Claire decides to take drastic measures.

With Lowery's help, Claire has Paddock Nine opened, unleashing the park's Tyrannosaurus rex, the elderly female that menaced the original Jurassic Park decades ago. Claire lures the elder rex to fight the Indominus. The old dinosaur is quickly overwhelmed by the larger, more dextrous Indominus, but just as she is about to die, Blue returns and attacks the Indominus for wiping out her pack. The elderly rex rejoins the battle, and with Blue distracting the Indominus from making serious attacks, the rex puts her brute force to good work. The pair successfully wear down the hybrid by pushing it back to the edge of the park's lagoon, but before round three commences, the Mosasaurus leaps from the water and grabs the Indominus, dragging it underwater to feast. When the rex looks to Blue, the fighting over, Blue is submissive, and the rex accepts this and the pair peacefully part ways. Owen releases Blue from his command.

Jurassic World is apparently abandoned to the animals. The tourists are evacuated to the mainland, where Zach and Gray are reunited with their parents. Claire and Owen decide to remain together.

On Isla Nublar, the Tyrannosaurus rex looks over the abandoned park, and roars, reclaiming her kingdom.
