    Mario 
The main hero of the series.

    Luigi 
Mario's brother.

    Peach 
The Princess of the Mushroom Kingdom.

    Daisy (spoilers) 
Peach's cousin.
  • Brainwashed and Crazy: Daisy is brainwashed by the Amulet of Krad into becoming the fifth boss.
  • Damsel in Distress: Eventually gets kidnapped by Tatanga, albeit only for the duration of Worlds 3, 4, and 5.

    Bowser 
Ruler of the Koopa Kingdom and arch-enemy of Mario, he nevertheless is forced to team up with him to get back his kingdom.
  • Enemy Mine: Bowser teams up with Mario after losing to him in order to tack back his army from Tatanga.
  • Even Evil Has Standards: Bowser may be a wannabe conqueror, but even he considers Tatanga brainwashing Daisy to fight Mario particularly low.

    Tatanga 
An alien returning from Super Mario Land who serves as the antagonist of the game. Powered by the Amulet of Krad, Tatanga aims to become lord of the universe.
  • Aliens Are Bastards: He is a purple alien and the Big Bad who wants to rule the universe.
  • Big Bad: He is the one who takes over the Mushroom Kingdom and wants to conquer the universe.
  • Evil Laugh: “Nyahaha!”
  • Final Boss: The last enemy faced by Mario.
  • Final-Exam Boss: The last phase of his fight has him use each of the six Pearls of Wisdom to attack, with six mini-phases in which he utilizes one pearl each.
  • Galactic Conqueror: His goal is to become ruler of the universe.
  • Giant Hands of Doom: In his final form, he sports these and uses them for some attacks.
  • King Mook: The leader of the regular alien enemies.
  • One-Winged Angel: Once his first two phases are beaten, he uses the six pearls to become a floating head with Giant Hands of Doom.
  • Pet the Dog: Tatanga is a would-be conqueror who does some nasty things, but he actually has one moment after the Wizen battle where he checks up on his advisor Wizen to see if he is okay, and only continues with his plans after Wizen confirms he is fine.
    Tatanga: Hey, Wizen, you okay? You don't look so good.
    Wizen: I will be fine. Just go ahead with your plans.
    Tatanga: Okay.
  • Screw This, I'm Outta Here: Once the player collects the Miracle Stones after beating the game, he immediately decides to cut his losses and leave once Valkalara is unsealed.
  • Sequential Boss: He is fought in three phases. First, he fights Mario on foot with several attacks thanks to the Amulet of Krad. Then he escapes on his ship and Mario must shoot him down similalrly to the final battle in Super Mario Land. Finally, he goes One-Winged Angel and uses each of the six pearls to attack Mario, with six-mini phases, one for each pearl.
  • Took a Level in Badass: Tatanga in Super Mario Land was somewhat of a wimp who only shows up in the end, needs his ship to fight, and is not all that intimidating. Super Mario Land 2: 6 Golden Coins even reduces him to an ordinary boss fight. In this game, thanks to the Amulet of Krad, Tatanga is able to pose a far greater threat, sports a snazzy military dictator-esque outfit, and has a more difficult boss fight with three phases.

    Wizen 
The trusted advisor to Tatanga.
  • Climax Boss: He is the boss of World 7 and fought at the top of Tatanga's HQ, with his own unique battle theme.
  • The Dragon: Wizen is Tatanga's advisor who helps him with his plans and is the penultimate boss.
  • Just Following Orders: Wizen seems to not want to fight Mario in World 7, but Tatanga ordered him to.
    Wizen: I'm sorry it has to end this way, but I cannot disobey Tatanga's direct orders.

    Valkalara (spoilers) 
The evil force that long ago attacked Tatanga's people until being sealed away within the Amulet of Krad.
  • Battle in the Rain: Once she's unsealed, the sunny atmosphere soon turns stormy and doesn't return to normal until after Valkalara is defeated. Once Peach comes out to talk to Mario, it's implied that they merely dismiss the event as a flash storm.
  • Bullet Hell: Most of her attacks are hard to dodge energy projectiles.
  • Generic Doomsday Villain: Due to her being a Greater-Scope Villain with very little screentime, all that is known about her is that she is an evil force who nearly destroyed Tatanga's homeworld and is sealed within the Amulet of Krad, being the source of its power. When unsealed, the most we get in personality is that she wants to destroy the Mushroom World. Exactly why she seeks to destroy planets is never explained.
  • Greater-Scope Villain: Valkalara is the evil force that ravaged Tatanga's home planet and was sealed away within the Amulet of Krad, which Tatanga uses for his plans, but remains sealed away for the duration of the game unless all eight Miracle Stones are collected.
  • Sealed Evil in a Can: The powerful evil spirit that nearly destroyed the home planet of Tatanga is sealed within the Amulet of Krad. The Miracle Stones are what break the seal.
  • Superboss: After beating Tatanga, if Mario finds the eight Miracle Stones, then an extra level on the world map is unlocked in which Valkalara is released from the Amulet of Krad and fights Mario.
  • Tennis Boss: One of her attacks is a pink orb that must be reflected back 3 times to break down her barrier and deal damage to her.
  • The Dreaded: From what little we learn of her, Tatanga has every reason to flee once she's accidently unsealed by the Miracle Stones.
  • Walking Spoiler: Her mere existence is not revealed until just before the Tatanga battle and reveals the truth of the Amulet of Krad.
