Anna-Maria Presa
Voiced by: Regina Regan
Anna-Maria is the second main character in this game and George's new love interest. She asks George to help her decipher a manuscript which has been in her family for many generations. The manuscript is said to reveal the location of a certain treasure. It is later revealed that she used to work for the Order of St. Michael as a nun. She stole some manuscripts in the library prior to her disappearance.- Beware the Nice Ones: She's nice and sweet towards George but she's the one who stole Maynard's money in Phoenix and then the manuscript in Vatican.
- Bookworm: She has a lot of books about chemicals in her apartment. Father Gregor mentions to George that she worked as a librarian during her days as a nun.
- Braids of Action: Her character model wears her hair back in a plait.
- Foreshadowing: Her rosary and the fireplace in her apartment has a label of "Memento Mori".
- Heroic Sacrifice: At the end of the game, she holds Mevlut at gunpoint to buy enough time for George to save Nico and stop the activation of the Ark. She gets shot for her efforts.
- I Want My Beloved to Be Happy: Despite tricking George at the beginning of the game, she helps him to save Nico from getting sacrificed to power up the Ark.
- Locked Out of the Loop: She spends half of the game imprisoned by the Order of St. Michael.
- Naughty Nun: She confesses to George that she never took her vows after all.
- Romantic False Lead: For George.
- Tritagonist: After George and Nico, she is the most plot-revelant character in the game.
- Unwitting Pawn: She cooperates with the Order because she believes that they will bring peace to mankind.
Vergil
Voiced by: Bob Golding
Vergil is an ex-con computer hacker and George's partner at Big Bros. Bail Bonds Agency. He provides George a hacking tool which is used several times throughout the game.- Reformed Criminal: He offhanded mentions he has "been in prison".
- What Happened to the Mouse?: After he teaches George to hack servers, he is never mentioned again.
Brother Mark
Voiced by: Wayne Forester
Brother Mark is a priest in training and Anna-Maria's neighbor. He has interest in action films and a big fan of Lucy Chu.- Ambiguously Gay: Until he mentions he wants a woman to see him in action.
- Big Damn Hero: He assists George in Vatican to rescue Nico.
- Closet Geek: He likes to watch action films and is a big fan of Lucy Chu.
- Conspicuous Gloves: He is introduced wearing a pair of hot pink gloves while he is sweeping the street in front of his apartment building.
- Cool Car: Mark named his yellow car after his favorite action star.
- Dissonant Serenity: When he and George discover there's a bomb in Anna-Maria's apartment, he tries to stay calm and trusts George that he can defuse the bomb.
- Hot-Blooded: He even mentions it after he helps George unlock the chamber.
- I Know Karate: He learned Tai Chi, ju-jitsu and Korean kick-boxing.
- Irony: Mark abhors violence, but he enjoys watching action movies.
- Neat Freak: He constantly cleans outside the apartment.
- Only One Name: He is only referenced by the title "Brother". Any surname is not given to the player.
- The Precious, Precious Car: He doesn't want anyone to mess with his car. George uses this as a distraction to enter Anna-Maria's apartment.
- Taking the Bullet: Near the end of the game, his right shoulder is shot by the evil monks after he calls the mafia. He survives and is glad that he finally experience being an action hero.
Archibald "Archie" Lonsdale
A homeless man who stays outside Anna-Maria's apartment building. He came from England to write a book about wines. He is easily distracted by women such as Nico.
- Abhorrent Admirer: To Nico.
- The Alcoholic: He always speaks with a slurred voice, and is seen with a bottle of an alcoholic beverage whenever George finds him.
- Big Damn Hero: He saves George and Anna-Maria in Vatican by knocking down the evil monk.
- Crazy Homeless People: He says he is Viscount Lonsdale, and mentions a military operation in Monte Cassino, in 1943, with General Leese againt Kesselring.
- Distracted by the Sexy: Done right after he sees Nico's back while talking to George.
- Grievous Bottley Harm: Archie knocks the monk's head using a wine bottle to save George and Anna-Maria.
- Hidden Depths: He keeps a stash of wine in the cellar at the monastery in Vatican.
Mevlut
Voiced by: Toby Longworth
A Turkish undercover secret agent. He is after George who has connections with mafia. Like George, Mevlut also investigates the disappearance of the ark.- Dirty Cop: He reappears at the very end of the game, in the Vatican catacombs, and exposits that, to him, finding the Ark is to sell it to the highest bidders.
- Good Smoking, Evil Smoking: According to a concept art.
- Heel–Face Revolving Door: Mevlut sets George free because George is looking for the Ark as well.
- Master of Disguise: He was the waiter who provided room service at the Pasha Palace, where Anna-Maria and George stayed while in Turkey.
- Only One Name: He is only known by the appellation Mevlut; either a name or a surname, the player never knows.
Dr. Ronald Maynard
The scientist who is assigned to make monatomic gold for the Order of St. Michael. He appears that he's wearing a headband and arms an AK-47. He is somewhat eccentric and admits to eating gold, which he believes protects him from danger.
- Crazy Survivalist: After his desert lab explodes, he uses an AK-47 to fend off invaders to the lab ruins.
- Extreme Omnivore: He believes that eating gold will protect him from harm.
- Happily Married: He uses his wedding anniversary as a password for his laboratory.
- Mad Scientist: He created the "Monoatomic Gold" - which appears to be inexistant in nature, due to being a physical impossibility - and gloats that he can use it to cure illnesses and prolong life.
- Taken for Granite: He is accidentally trapped in a machine and turns into gold.
- Trigger-Happy: He loves guns, especially A.K.-47.
Alfonso
Voiced by: Toby Longworth
The owner of a shabby hotel in New York. He has problems in getting rid of gangsters.- Tragic Keepsake: He has the golden lighter from his father who died in a gas explosion.
- The Stoic: He isn't amused at the state of his hotel.
Thelwell Minster
A middle-aged man who gets tricked by Alfonso's advertisement.
- Friend to All Living Things: He likes geese.
Officer O'Halloran
A policeman in New York who investigates Alfonso's hotel and its connection to the mafia.
Juanita
Voiced by: Regina Regan
Alfonso's sole receptionist and cleaner who is kidnapped by the mafia.- Bound and Gagged: She is trapped in the closet by the mafia.
- The Tease: After George goes back to Alfonso's hotel and inquires Juanita, she flirts a bit with George (compliment his hands, asking him to see the bump on her head).
Toni Martino
Voiced by: Bob Golding
One of the members of mafia who occupied the hotel. He's the smart member of the group and has a business degree.- You Have Failed Me: His body is hung in the factory.
Fingers Martino
The big guy of the group and somewhat dimwitted.
- Trademark Favorite Food: He loves salami.
Chico Garella
The oldest member of the mafia. He acts as a seller in a salami shop to cover their crimes.
- Catchphrase: "Yeap! It's a screw-up alright!"
- Trademark Favorite Food: He likes salami.
Vincenzo Spallacci
Spallacci is the godfather of the Martino crime family. He orders his members to chase Anna-Maria and steal the manuscript.
- Closet Geek: Like Brother Mark, Spallacci has an interest in action movies and a bigger fan of Lucy Chu.
- Good All Along: He cooperates with Cardinal Gianelli in stopping the order of St. Michael.
- Heel–Face Turn: He reveals that Cardinal Gianelli changed his life and made him realize his evil actions.
- Never Hurt an Innocent: He tells Fingers via radio that he shouldn't kill Nico, who knows nothing about the incident regarding the Order of St. Michael.
- The Leader: Of the Martino family mafia.
- Suspiciously Similar Substitute: To Khan.
Cardinal Gianelli
A member of the Holy Order who sends the mafia after Anna Maria to retrieve the manuscript she stole.
- Dissonant Serenity: Even he faces Father Gregor with a gun, he manages to keep calm.
- Good All Along: He's trying to stop Father Devlin and Father Gregor from using Ark to kill innocents.
- I'm Dying, Please Take My MacGuffin: At Vatican, he gives the manuscript to George and Brother Mark before he dies.
- Perpetual Frowner: He always looks serious.

- Wall Slump: He dies in this position after he tells George and Brother Mark to stop the Ark.
Monsignor Devlin
Member of the Holy Order who gives information about Anna-Maria to George.
- Villain: Exit, Stage Left: He disappears along with the two monks near the end of the game.
- Well-Intentioned Extremist: His order's mission is to bring faith to millions of people by using the Ark to destroy the non-believers.

He is the assistant of Mosignor Devlin who hired Anna-Maria to find the Ark's missing cherubs.
