- Channel Hop: As with any DreamWorks Animation franchise that began between 2013 and 2017, Boss Baby switches distributors from Twentieth Century Fox to Universal. The film's availability on Peacock is also a switch from The Boss Baby: Back in Business, which aired on Netflix (where the first film also made its streaming premiere).
- The Other Darrin:
- James Marsden replaces Tobey Maguire as the voice of adult Tim. Marsden also voices Tim in his child form after he takes the new formula, replacing child actor Miles Bakshi, who voiced child Tim in the original film.
- Ariana Greenblatt replaces Nina Zoe Bakshi as Tim's eldest daughter Tabitha while also reprising the role from the The Boss Baby: Back in Business.
- Role Reprise: Overlapping with The Other Darrin, Ariana Greenblatt reprises Tabitha from The Boss Baby: Back in Business.
- Release Date Change: It was going to be released in March of 2021, but COVID-19 caused it to be delayed to September of that year before settling for July.
