• The Original Darrin: Yuri Lowenthal returns to voice the title character after Robin Atkin Downes voiced him in the previous game.
• The Other Darrin: The old man is the only character who kept the same voice actor from the previous game.
  • Monica Bellucci who voiced Kaileena in Warrior Within is replaced by Sarah Carlsen.
  • Joanna Wasick who voiced Farah in Sands of Time is replaced by Helen King.
  • Barry Dennen who voiced the Vizier in Sands of Time is replaced by Harry Standjofski.
  • Michael Rudder voiced the Dark Prince in the ending of Warrior Within. Here, he's voiced by Rick Miller.
• Urban Legend of Zelda: No matter what you do, The Two Thrones only has one ending. There is no bad ending where the Dark Prince wins. This was mostly fueled by the fact that the previous game (Warrior Within) had two endings, and the Golden Ending shows an Early-Bird Cameo of the Dark Prince.
