Hikaru Tsuki is a new 5th grade teacher at Tozai Elementary School. Secretly, she is also the alien princess of Planet Alpha and a peacekeeper for the Space Alliance assigned to Earth.
The first 13 episodes are mainly comedy-focused where Hikaru would use her magical powers to help her students with mundane issues. Midway through however, Hikaru gained the power to become a superheroine named "Andro Mask", resulting in Majo Sensei being the first Toku show to feature a female Henshin Hero. From then on the show became more action-focused, with Hikaru repeatedly doing battle against a recurring Arch-Enemy in the form of the blood-sucking alien Kumondes.

- Action Girl: Hikaru. Even before she becomes Andro Mask, she shows multiple times that she's capable of handling herself in a fight.
- By the Power of Grayskull!: Hikaru always chants "Moonlight Power" to use her abilities. To transform into Andro Mask, she chants "Andro Kamen, ro!"
- Cool Teacher: Hikaru, who is well-liked by her students for her very unorthodox teaching style.
- Distinguishing Mark: Episode 20 features Rumi, a girl with a rather large mole on the left side of her face that makes her the target of teasing.
- Dream Episode: Episode 4 has Hikaru traveling inside of Susumu's dream to beat up his night terror so he can get a good night's sleep.
- Education Mama: Susumu's mother, whose constant insistance on making him study eventually causes him to develop night terrors. Episode 14 sees her become even worse under the influence of Kaijin Dictionary, who coerces her into locking Susumu in his room to make him study and put bars on his window so he can't escape.
- Halfway Plot Switch: The first half is an outright comedy. The second half sees the series transition from light-hearted fun into an actionized Henshin Hero show.
- Henshin Hero: Hikaru, once she gains the power to transform into Andro Mask.
- Hot Teacher: Hikaru obviously, who is played by a 21 year old Yoko Kiku. Her male counterpart, Teacher Hatano, is also quite handsome to the point where Hikaru herself even begins to develop a bit of a crush on him.
- Knight of Cerebus: While the show was initially full of light-hearted comedy and even after Hikaru became Andro Mask most plotlines weren't all that serious, Kumondes singlehandedly disrupts the fun with his introduction and repeated plans to drink the blood of Hikaru's students.
- Non-Human Sidekick: Baru. Hikaru isn't human herself, but Baru doesn't even look human, instead being an old, rabbit-like alien.
- Sensei-chan: Hikaru is an early example in Japanese media. She's friendly, popular with her students and attractive, though she doesn't care as much about being single as other examples.
- Zettai Ryouiki: From episode 12 onward, Hikaru starts wearing a new outfit that trades out her white heels for a pair of black, thigh-high boots.
