Wade Wilson, the loudmouth mercenary known as "Deadpool", is having a very bad day. On top of Wolverine having the audacity to up and die in his final movie, his life as a murderous soldier-of-fortune catches up to him when, on his anniversary with his lover Vanessa, a group of criminals breaks into their home, one of them shooting and killing Vanessa. Blaming himself for her death, he spends the next six weeks trying to end his life: throwing himself into a polar bear pit at the zoo, leaping off a building after chugging drain cleaner, and finally blowing himself up while lying on some barrels of explosive chemicals.

After the attempted death-by-explosion, Wade has a vision of Vanessa telling him, "Your heart's not in the right place..."

Wade awakens in Xavier Mansion, his body having been recovered by Colossus. Colossus once more asks Wade to join the X-Men, and, believing this is what Vanessa wanted, Wade accepts. His first mission sees him accompany Colossus and Negasonic Teenage Warhead to an asylum where young mutants are "cured" via torture, where one of the inmates, a young boy named Russell Collins — aka "Firefist" — is having an armed standoff with the police. Recognizing Russell as a victim of abuse at the hands of the asylum's staff, he shoots and kills one of the nurses before being restrained by a disappointed Colossus. Wade and Russell are both arrested, outfitted with power-suppressing collars, and sent to a prison for mutants — the "Ice Box".

As the collar suppresses his healing factor and causes cancer to rear its ugly head, Wade, believing himself not long for this world, tries to distance himself from Russell, to the young boy's distress. Unfortunately, his troubles are only getting worse: a time-traveling cyborg mutant known only as "Cable" breaks into the Ice Box to hunt down and kill Russell. Wade comes to the boy's defense, but claims not to care for him, breaking Russell's heart as the two fighting mutants fall out of the prison's walls. Wade has another vision where Vanessa tells him that he must help Russell...

As Russell plots and plans his revenge along with a friend he made in the Ice Box, a massive and powerful mutant known as the "Juggernaut", Wade meets with his friend Weasel to gather a group of mutant mercenaries to help him save Russell. Answering his call to arms are Bedlam, a mutant with the power to manipulate electromagnetic fields to interfere with machines and human brains; Shatterstar, an alien who claims to be better than humans at everything; Zeitgeist, a mutant with the ability to vomit acid; Vanisher, a mutant with the ability to turn invisible; Domino, a mutant with exceptionally good luck; and Peter, a regular human being with no outstanding abilities.

Christening his new squad the "X-Force", Wade and his posse attempt to intercept a prison convoy transporting Russell via air drop. Unfortunately, the attempted air drop goes horrifically awry; Bedlam is hit by a bus, Shatterstar gets pulped by a helicopter's blades, Zeitgeist falls into a woodchipper, Peter dies trying to save Zeitgeist when he gets hit with acidic vomit, and Vanisher gets electrocuted on power lines. Only Wade and Domino survive the air drop. While Wade and Domino fend off Cable once more, Russell frees himself and the Juggernaut, who tears Wade in half, before escaping to help Russell get his revenge against the asylum's director.

While recovering from his run-in with the Juggernaut, Wade is approached by Cable, who explains why he is trying to kill Russell: in the future he hails from, Cable's wife and child were murdered by Russell, who developed a bloodlust after killing the asylum's director and went on to become a serial murderer with his pyrokinetic abilities. Cable came back in time to kill Russell before he could turn into a murderer in the hopes of changing history so his family survives. Wade asks Cable for a chance to try and convince Russell to abandon his desire to kill, which Cable reluctantly grants.

Together, Wade, Cable, Colossus, Negasonic Teenage Warhead, and fellow X-Man Yukio make their way to the asylum. While Colossus and his compatriots deal with the Juggernaut, Wade and Cable find Russell, who is about to kill the director, and Cable prepares to shoot the boy. Wade, in a last-ditch effort to save Russell from himself, places a power-suppressing collar on himself and dives in the path of Cable's gun, taking a fatal bullet. Russell, shocked and moved by Wade's sacrifice, gives up his pursuit of revenge, changing the future and saving Cable's family...

Wade and Vanessa briefly reunite in the hereafter, but Vanessa tells him it's not his time yet: also moved by Wade's sacrifice, Cable uses the last of his time travel device's energy to go back to when he and Wade arrived at the asylum, this time leaving Wade's anniversary gift from Vanessa, a lead skee-ball token, in his suit to block the bullet. With Russell's desire for vengeance abandoned and Cable's family still safe (plus the asylum director dying anyway shortly after thanks to a hit-and-run), Wade and his friends live happily ever after...

Not only that, but with the help of Negasonic Teenage Warhead and Yukio, Wade manages to repower Cable's time travel device and use it to set right the wrongs of the past: saving Vanessa from her death, convincing Peter to walk away from X-Force before Fox's desire to keep Josh Brolin employed kills him, putting Weapon XI out of his misery while giving Wolverine some sage advice, stopping Ryan Reynolds from making the biggest mistake of his career, and finally, killing Baby Hitler...note
