Definitely not Lupus is a crossover fanfic between Once Upon a Time and House. It tells the story of House and his team getting stuck in Storybrooke where they run into an old friend, Dr. Allison Cameron, who is revealed to be Emma Swan using a fake identity. Meanwhile, Regina struggles to keep them from discovering the truth and keep Emma, now the Dark One, under control.

- Big Bad: The essence of the Dark One is behind Belle trapping House's team there, so that they can sacrifice Chase, Emma, and Regina to be free of Emma before it dies.
- Butt-Monkey: Chase is repeatedly beaten down for the sake of comedy. House is constantly mocking him over Cameron (Emma) being married to Regina, who he sees as trading up. Snow hits him with a lamp for breaking Emma's heart. Emma knocks him out with a clock and Regina punches him for suggesting that Emma would cheat on her. Emma pretends to seduce him and then rips his heart out.

When Foreman and Thirteen even try to suggest that Regina abuses her, Emma shuts it down immediately, and even as the Dark One, Emma refuses to let anyone insult Regina, as evidenced when she nearly chokes Chase to death for suggesting it. Also, when Regina notices that Emma is still attracted to House, she doesn't get jealous, as she is secure that Emma would never cheat on her.
- Mistaken for Cheating: Subverted. When everyone finds out that Emma is pregnant, Chase attempts to claim that Emma didn't tell Regina because she obliviously cheated on her. Regina doesn't buy into it for a second and instead simply punches Chase for even suggesting it.
- Moment Killer: After Emma manages to convince Regina to kiss her, Chase walks in on them making out and instantly kills the mood. For doing so, Emma knocks him out with a clock.
- Mystical Pregnancy: Emma is revealed to be pregnant with Regina's baby. This is explained as their true love creating it and everyone is shocked that this could happen.
- One True Love: Emma and Regina are constantly called true loves throughout the story. Their love is strong enough that Emma becomes pregnant through said love, and when they kiss, it manages to give Emma her magic back.
- Trademark Favorite Food: In one scene in the story, Emma requests that Regina bring her something other than hospital food. Regina says she assumed that Emma was a fan of food in general, to which Emma replies that her favorite food is grilled cheese.
Regina: I thought you to be a fan of all food.
Emma: I'm the number one fan of grilled cheese.

- Unresolved Sexual Tension: Regina quickly picks up on the fact that Emma and House are attracted to each other despite not seeing each other for years and their constant bickering. Strangely she doesn't mind it, as she is positive that Emma would never cheat on her.
