Andrew and Jake Basher are brothers who run their dead father's garage in Kern County. Despite being pretty competent mechanics, Andrew's girlfriend Vanessa left him because he wouldn't "grow up" for Dennis, the CEO of a subsidiary for Tronco with an office located in Kern County.
One night, after a visit to the bar Vanessa works at, Jake is attacked by a giant monster that bites his arm. Andrew manages to save Jake, but Jake's arm has been transformed into a monster arm as a result. Soon, however, Kern County finds itself experiencing a rash of attacks by monsters.
Now, Andrew, Jake, with the help of Vanessa need to get to the bottom of this mystery, assuming the monsters don't kill them all first.

We Kill Monsters contains examples of:
- Alien Blood: The first of the monsters that Andrew killed had blue blood. The giant insect monster had green blood.
- Big Creepy-Crawlies: Vanessa's father is kidnapped and killed by a giant insect monster.
- Corporate Conspiracy: Tronco created the monsters via genetic engineering and plans to integrate their DNA into all the products of the companies they own and sell it to humanity to integrate it into their bodies. The apparent aim is to make humanity much stronger and healthier with the monster DNA.
- Extra Eyes: The first kind of monster that Jake and Andrew encounter has three eyes.
- Eye Scream: When Andrew drives a shovel into the skull of the monster attacking Jake, he drives it into where the third eye appears to be located.
- Fat and Skinny: Jake and Andrew, respectively. Jake even comments on his appearance when he sees himself through the eye on his newly transformed tentacle arm.Jake: Hey, whoa, I'm kind of fat.
- Genetic Abomination: All the monsters are genetically modified creatures created by Tronco for their Corporate Conspiracy.
- Harpoon Gun: Jake comes up with the idea of making a large harpoon gun mounted on their truck to catch and bring down the giant insect monster that killed Vanessa's dad.
- Shovel Strike: Andrew saves Jake from the monster that attacked him by driving a shovel into the creature's skull.
- You Are Number 6: Andrew finds a serial number on the remains of the bug monster he, Jake, and Vanessa killed, reading M87009-3XR. It leads him to deduce what the true origins of the monsters is.
- You Killed My Father: Vanessa joins Andrew and Jake on their mission to kill the giant insect monster that took her father before her very eyes.
