Mario
- Adaptational Jerkass: Downplayed. While he's still as selfless and courageous as his canon self, he's also more cynical and sarcastic. And even as the series goes on, he gets better.
- Character Catchphrase: "Let's-a-go" and "Mamma Mia," naturally. Ironically, "It's-a-me, Mario!" is never said until Doopliss impersonates him in Season 1's penultimate episode.
- Playing with Fire: He slowly develops his Firebrand abilities over the course of the series.
Luigi
- Shock and Awe: In "The Comeback," he begins to develop his Thunderhand abilities.
Peach
Daisy
Link
- Chick Magnet: When the homecoming dance rolls around, mobs of girls (and Byleth) chase after him. It gets to a point where he needs Zelda's help to hide.
Zelda
Wario
Waluigi
Rosalina
Donkey Kong
- Lovable Jock: He's a member of Nintendo High's football team, and is nothing but friendly to Mario and his pals. He's even the first friend Mario makes in the series premiere.
K. Rool
Falco
Slippy
Toad
Pit
Dark Pit
Captain Falcon
- Butt-Monkey: He is often subjected to this. He gets brutally rejected by Daisy, his Falcon Punch isn't nearly as powerful as in Smash, Samus nearly tramples him with a Tauros, and his real name is "Douglas."
- Those Two Guys: He and Samus are often seen together.
Samus
- Those Two Guys: She and Captain Falcon are often seen together.
Kirby
Byleth
Claude
- Adaptational Sexuality: He attends the Homecoming Dance Byleth, who is canonically bisexual. While Fire Emblem: Three Houses has a hefty amount of Gay Options, Claude wasn't one of them.
Francis
Goron
Antagonists
Bowser
- Big Bad: For the first season, naturally.
Mouser
Nastasia
O'Chunks
Mimi
Dimentio
Doopliss
Pauline
- Adaptational Jerkass: In the games, Pauline is portrayed as being down-to-earth and helpful. Here, she's an over-the-top Diva competing for Mario's heart.
- Adaptational Villainy: Unlike in her home series, Pauline is depicted as a rival to Peach, often competing with her for Mario's attention.
Faculty
Master Hand
- Horrible Judge of Character: He somehow thinks Mario and Luigi are bullying Wario and Waluigi, instead of the other way around.
Professor E. Gadd
- Age Lift: While this is normal for a High School AU, E. Gadd is a special case, since he's still an adult. But since his hair is brown, implying that he's younger than he is in the games.
Tingle
Nurse Joy
Doc Louis

Students
Sonic
- New Transfer Student: After he got expelled from Sega High, he switched to Nintendo High. But it's later revealed that he wasn't expelled. He left because he couldn't handle the pressure of being The Hero of the school.
Tails
Knuckles
- Tsundere: While he'll never admit it, he's just as bummed as Tails after Sonic leaves Sega High. After finding the blue blur at Nintendo High, he claims in only came because Tails wouldn't shut up about him.

Dr. Eggman
