The second trailer shows them as two antagonists looking for Betelgeuse. One theory is that they are a couple like the Maitlands were, only they didn't avoid getting screwed over by him the way Adam and Barbara did, which is why they look worse for wear.

As she said that as her character is a ghost is not supposed to age, but since she has she can't return (and wont') as they would have to explain why and how ghost aged 35 years. Dafoe's character in the film is after Beetlejuice is formerly the assistant of Juno (the Maitland's case worker from the first film) who replaced Beetlejuice after he became a "bio-exorcist." Before Juno retired, she tasked him to keep an eye on Beetlejuice to make sure he didn't get back out and cause more chaos. This explains why Dafoe is screaming at the shrunken head guy in the trailer, because Beetlejuice used him as a BodyDouble for his escape (this explains why the shrunken head guy is dressed in Beetlejuice's iconic black and white striped suit and sports a similar hairstyle). Dafoe's character is looking for Beetlejuice to keep him secure from the land of the living.
