* TheStinger of the Film/2016 reboot indicates that the original film's Zuul, and possibly by extension Gozer, would be the main villain of a sequel. However, due to the film's underperformance, said sequel never materialized and the Unreboot Film/GhostbustersAfterlife was developed instead.

* Film/KamenRiderTheFirst zig-zags this. While the typical NebulousEvilOrganisation Shocker appears, the Great Leader doesn't make an appearance until the sequel, Film/KamenRiderTheNext. But he's still mostly a GreaterScopeVillain, with the more immediate threat being Shiro Kazami's mutated sister.

Though Kazami himself as Kamen Rider V3 could also count as an iconic character initially serving as an antagonist.

* Feyd-Rautha Harkonnen is a fairly iconic character of Literature/Dune and didn't appear in Dune: Part One, instead getting an AdaptationalLateAppearance in Film/DunePartTwo (though the two parts still adapt the first novel anyway).

*** In Film/2018, Black Manta features in a subplot detailing his origin and hatred of Aquaman, culminating in a skirmish between the two with Manta in full costume. However, the main threat is the less-iconic Ocean Master, with TheStinger seemingly setting up Manta as the primary villain of the sequel. Ultimately, said sequel, Film/AquamanAndTheLostKingdom, does feature Manta as the primary physical threat for Aquaman, but he spends most of the film possessed by SealedEvilInACan Karthax, who uses Manta to attempt to free himself so he can wreak havoc on the world.
